shBclaf1 is a short hairpin RNA (shRNA) designed to suppress the expression of BCLAF1 (BCL2-associated transcription factor 1) through RNA interference (RNAi). BCLAF1 is a transcriptional regulator that plays a critical role in hematopoietic stem cell (HSC) self-renewal and is often upregulated in pediatric and adult acute myelogenous leukemia (AML). In research settings, shBclaf1 is used to demonstrate that the suppression of BCLAF1 is detrimental to AML cell survival and pathogenesis. Studies have shown that AML cells expressing shBclaf1 are selectively depleted, and the loss of BCLAF1 leads to a dramatically altered genetic program associated with metabolic, apoptotic, and myeloid differentiation pathways. This tool has been utilized in preclinical research at the Washington University School of Medicine to investigate BCLAF1 as a potential therapeutic target in leukemia.
