shBRD4 is a short hairpin RNA (shRNA) designed to silence the expression of Bromodomain-containing protein 4 (BRD4), a member of the BET (bromodomain and extra-terminal motif) family. BRD4 acts as an epigenetic reader that binds to acetylated lysine residues on histones, playing a critical role in regulating the transcription of genes involved in cell cycle progression and proliferation. In preclinical studies, particularly in melanoma, shBRD4 has been used to demonstrate that BRD4 knockdown suppresses tumor cell growth, colony formation, and in vivo tumor progression. This research tool helped validate BRD4 as a therapeutic target, leading to the development of small-molecule BET inhibitors.
