Overview

shCREB refers to a short hairpin RNA (shRNA) construct designed to silence the expression of the cyclic AMP response element binding protein (CREB), a transcription factor that plays a critical role in cell survival, proliferation, and drug resistance. In cancers such as malignant mesothelioma (MM), CREB is often constitutively activated, driving the expression of pro-survival genes like Bcl-2 and Bcl-xL and promoting the secretion of angiogenic and inflammatory cytokines (e.g., VEGF, IL-6, IL-8). By knocking down CREB expression, shCREB attenuates tumor growth, induces apoptosis, and enhances the sensitivity of cancer cells to chemotherapeutic agents like doxorubicin. This approach is primarily utilized in preclinical research to investigate the therapeutic potential of targeting the CREB signaling pathway.
