Overview

shDSG3 is a short hairpin RNA (shRNA) designed to silence the expression of Desmoglein 3 (DSG3), a protein that acts as an oncogene in head and neck cancers (HNCs). By suppressing DSG3, the drug disrupts the interaction between DSG3 and plakoglobin, leading to the translocation of plakoglobin from the cytoplasm to the nucleus. This translocation inhibits TCF/LEF transcriptional activity, subsequently downregulating downstream oncogenic targets such as c-myc, cyclin D1, and MMP7. Research conducted at Chang Gung University has demonstrated that shDSG3 reduces cell growth, colony formation, migration, and invasion in HNC cell lines and inhibits tumor growth in xenograft mouse models.
