Overview

shEZH2 is a short hairpin RNA (shRNA) construct designed to silence the expression of the Enhancer of Zeste Homolog 2 (EZH2) gene through RNA interference (RNAi). EZH2 is the catalytic subunit of the Polycomb Repressive Complex 2 (PRC2) and functions as a histone methyltransferase that methylates lysine 27 of histone H3 (H3K27), leading to transcriptional repression. In oncology research, particularly in glioblastoma multiforme (GBM), EZH2 is identified as a key regulator of glioblastoma stem cell (GSC) maintenance and tumor progression. shEZH2 is utilized as a targeted tool to downregulate EZH2 expression, thereby inhibiting downstream signaling pathways such as STAT3 activation. Preclinical studies, including those conducted at the Cleveland Clinic, have demonstrated that shEZH2-mediated knockdown can impair GSC self-renewal and, when combined with other inhibitors, extend survival in animal models of intracranial tumors.
