Shigella flexneri 2a-rEPAsucc is a glycoconjugate vaccine candidate developed to prevent shigellosis caused by *Shigella flexneri* serotype 2a. The vaccine consists of the O-specific polysaccharide (O-SP) of *S. flexneri* 2a covalently linked to a recombinant Pseudomonas aeruginosa exoprotein A (rEPA) carrier protein. The 'succ' designation indicates the use of a succinic acid derivative (specifically succinic acid dihydrazide) as a linker or a succinylation modification to facilitate the conjugation of the polysaccharide to the protein carrier. This conjugation transforms the T-cell independent O-antigen into a T-cell dependent antigen, thereby enhancing immunogenicity and inducing long-lasting immunological memory. It was primarily developed by researchers at the National Institutes of Health (NIH) and has been evaluated in clinical trials for its safety and ability to elicit serum IgG antibodies against the Shigella lipopolysaccharide.
