The Shigella sonnei O-SP-core conjugate vaccine (O-SPC-rDT) is an investigational glycoconjugate vaccine developed by the Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD) for the prevention of shigellosis. The vaccine consists of a fragment of the *Shigella sonnei* lipopolysaccharide (LPS), comprising the O-specific polysaccharide (O-SP) and the core region (O-SPC), which is covalently bound to a recombinant non-toxic diphtheria toxin mutant (rDT) carrier protein. The saccharide component is designed with a low molecular mass, averaging 3.5 repeat units, and is coupled via its reducing end to enhance immunogenicity compared to previous full-length O-SP conjugates. The vaccine's mechanism of action involves eliciting type-specific serum IgG antibodies against the O-SP domain, an essential virulence factor and protective antigen of *S. sonnei*. It is currently being evaluated in Phase 1 clinical trials to assess safety and immunogenicity in healthy adults, with the ultimate goal of providing protection to young children, the population most affected by shigellosis.
