Overview

Shigella sonnei strain 53G is a well-characterized laboratory and clinical challenge strain of the Gram-negative bacterium *Shigella sonnei*, which is a major cause of bacillary dysentery (shigellosis) worldwide[1][3][5]. Strain 53G belongs to lineage II and has been widely used in controlled human infection models (CHIMs) for vaccine development and pathogenesis studies[1][3][6]. The organism possesses a virulence plasmid (pINV) encoding a type III secretion system essential for host cell invasion and disease manifestation[2]. Lyophilized preparations of this specific strain have been produced under current good manufacturing practice (cGMP) standards for use in standardized human challenge studies[3][6]. Infection with this challenge organism reliably induces shigellosis symptoms such as diarrhea, fever, and abdominal pain in healthy adult volunteers at defined doses; it serves as an important tool to evaluate candidate vaccines and study host-pathogen interactions[5][8].
