Overview

WRSS1 is a live attenuated, oral vaccine candidate developed by the Walter Reed Army Institute of Research (WRAIR) to protect against *Shigella sonnei* infection. The vaccine strain is derived from the wild-type *S. sonnei* strain Mosely and is attenuated through the deletion of the *virG* (also known as *icsA*) gene, which is essential for intra- and intercellular bacterial spread, as well as the *sen* and *set* genes that encode Shigella enterotoxins 1 and 2. By mimicking a natural infection without causing clinical shigellosis, WRSS1 aims to elicit protective mucosal and systemic immune responses. Clinical development has included Phase 1 trials in both adult populations and pediatric cohorts in endemic regions like Bangladesh and Thailand, supported by organizations such as PATH and the U.S. Army Medical Research and Development Command.
