Drug intelligence / Profile preview
Shigella4V (also known as S4V or S4V2) is a tetravalent bioconjugate vaccine candidate designed to prevent shigellosis, a severe and potentially life-threatening diarrheal disease caused by Gram-negative *Shigella* bacteria. Developed by LimmaTech Biologics and licensed to Valneva, the vaccine targets the four most common pathogenic *Shigella* serotypes: *Shigella flexneri* 2a, 3a, 6, and *Shigella sonnei*, which are responsible for up to 85% of global shigellosis cases. The vaccine is produced using bioconjugation technology in engineered *Escherichia coli*, where the O-antigen polysaccharides of the target *Shigella* strains are enzymatically linked to a genetically detoxified Pseudomonas aeruginosa exotoxin A (rEPA) carrier protein. Shigella4V has received Fast Track designation from the U.S. FDA and is currently being evaluated in Phase 2 clinical trials, including pediatric safety and immunogenicity studies in low- and middle-income countries (LMICs) and controlled human infection model (CHIM) trials.
