Overview

SHIP1 inhibitors are small molecule therapeutic candidates designed to block the activity of Src homology 2-containing inositol 5'-phosphatase 1 (SHIP1). SHIP1 is a signaling enzyme that negatively regulates the PI3K pathway by dephosphorylating phosphatidylinositol 3,4,5-trisphosphate (PI(3,4,5)P3) into phosphatidylinositol 3,4-bisphosphate (PI(3,4)P2). By inhibiting SHIP1, these compounds increase cellular levels of PI(3,4,5)P3, thereby enhancing PI3K-mediated signaling. While SHIP1 activators are typically developed for anti-inflammatory indications, SHIP1 inhibitors are primarily investigated for their potential to enhance anti-tumor immune responses or to treat specific hematological malignancies where SHIP1 activity may be detrimental to therapeutic outcomes. Aquinox Pharmaceuticals previously listed a SHIP1 inhibitor program in its preclinical pipeline, exploring the modulation of these signaling pathways for oncology and immune-related applications.
