shMCT4 is an experimental short hairpin RNA (shRNA) designed to silence the expression of Monocarboxylate Transporter 4 (MCT4), also known as SLC16A3. MCT4 is a key protein involved in the efflux of lactate from glycolytic cells, and its expression is often upregulated in response to hypoxia in various cancers, including glioblastoma. By reducing MCT4 levels, shMCT4 inhibits tumor cell growth and survival, particularly within the stem-like CD133-positive cell population. Research from Johns Hopkins Medical Institutions has shown that MCT4 knockdown via shRNA significantly reduces the clonogenicity of glioblastoma neurospheres and slows the growth of intracranial xenografts in vivo, suggesting that MCT4 is a viable therapeutic target for aggressive brain tumors.
