Overview

SHMDA-9 is an experimental short hairpin RNA (shRNA) therapeutic candidate designed to silence the expression of Melanoma Differentiation Associated gene-9 (MDA-9), also known as syntenin-1 or SDCBP. MDA-9 is a scaffolding protein that facilitates the assembly of signaling complexes involved in cancer cell motility, invasion, and angiogenesis. By utilizing the RNA interference (RNAi) pathway, SHMDA-9 mediates the degradation of MDA-9 mRNA, which in turn suppresses the activation of downstream effectors such as c-Src, p38 MAPK, and NF-κB. Preclinical studies in glioblastoma multiforme (GBM) models have demonstrated that SHMDA-9-mediated knockdown reduces tumor cell invasiveness, inhibits the secretion of angiogenic cytokines like IL-8, and slows tumor growth in vivo.
