Overview

shMetadherin is a short hairpin RNA (shRNA) construct designed to silence the expression of the Metadherin (MTDH) gene, also known as Astrocyte Elevated Gene-1 (AEG-1). Metadherin is a well-characterized oncoprotein that promotes tumor progression, metastasis, and chemoresistance in various cancers, including breast cancer. By utilizing the RNA interference (RNAi) pathway, shMetadherin mediates the sequence-specific degradation of MTDH mRNA, resulting in a significant reduction of MTDH protein levels. This knockdown inhibits downstream oncogenic signaling pathways such as PI3K/Akt, NF-κB, and Wnt/β-catenin, leading to decreased cell proliferation, migration, and invasion. In the context of breast cancer research, shMetadherin is frequently employed as a tool to investigate the role of MTDH in the tumor immune microenvironment and to validate MTDH as a potential therapeutic target.
