shNEAT1a is a short hairpin RNA (shRNA) construct designed to silence the expression of the long noncoding RNA (lncRNA) Nuclear Paraspeckle Assembly Transcript 1 (NEAT1). In the context of hepatocellular carcinoma (HCC), NEAT1 has been identified as a key mediator of resistance to multi-kinase inhibitors such as sorafenib and lenvatinib. Mechanistically, shNEAT1a-mediated knockdown of NEAT1 disrupts the NEAT1-SOD2 axis, leading to the suppression of AKT signaling and the activation of the MAPK/ERK pathway. This molecular shift sensitizes liver cancer cells to targeted therapies, suggesting that NEAT1 inhibition could be a viable strategy to overcome drug resistance in liver cancer patients. While currently utilized primarily in preclinical research, it represents a targeted RNA-based approach to modulating oncogenic signaling pathways.
