Shoc2 SMP Disruptor is an early-stage small molecule developed by SyntheX Labs designed to disrupt the protein-protein interaction (PPI) complex consisting of Shoc2, MRas, and the catalytic subunit of protein phosphatase 1 (PP1c). This ternary complex serves as a critical scaffold in the Ras/MAPK signaling pathway, facilitating the dephosphorylation and activation of RAF kinases. By interfering with the assembly of this complex, the drug aims to inhibit oncogenic signaling in Ras-mutated tumors, addressing a key node of oncogene addiction and synthetic lethality. The program is currently in the Hit and Mechanism of Action (MoA) validation phase.
