Overview

Short hairpin RNA targeting iASPP (sh-iASPP) is an RNA interference (RNAi) therapeutic candidate designed to silence the expression of the Inhibitor of Apoptosis-Stimulating Protein of p53 (iASPP), also known as PPP1R13L. iASPP is a key negative regulator of the p53 tumor suppressor family, binding to p53, p63, and p73 to inhibit their pro-apoptotic functions. In various malignancies, particularly cervical cancer, iASPP is frequently overexpressed, driving the epithelial-mesenchymal transition (EMT) and conferring resistance to platinum-based chemotherapies like cisplatin. By utilizing shRNA to knock down iASPP levels, this approach restores p53-mediated apoptotic pathways and modulates downstream signaling axes, such as the miR-20a-FBXL5/BTG3 pathway, to suppress tumor progression and enhance chemosensitivity. Currently, sh-iASPP is primarily utilized in preclinical research settings to investigate the molecular mechanisms of oncogenesis and potential gene therapy applications.
