Overview

Short hairpin RNA (shRNA) targeting PEBP4 is an experimental gene-silencing therapeutic designed to inhibit the expression of Phosphatidylethanolamine-binding protein 4 (PEBP4). PEBP4 is a multifunctional protein that is frequently overexpressed in various malignancies, including hepatocellular carcinoma (HCC), where it acts as a scaffold or regulator to promote tumor cell proliferation, migration, and invasion. By utilizing the RNA interference (RNAi) pathway, this shRNA construct triggers the degradation of PEBP4 mRNA, leading to a significant reduction in PEBP4 protein levels. This knockdown results in the suppression of the Akt/mTOR signaling axis, specifically inhibiting the activities of both mTORC1 and mTORC2. Preclinical studies have demonstrated that silencing PEBP4 with shRNA can effectively diminish tumor growth and metastasis in vivo, making it a potential candidate for targeted cancer therapy.
