Overview

This short hairpin RNA (shRNA) is an RNA interference (RNAi) therapeutic candidate designed to target and silence the *SLC9A1* gene, which encodes the sodium-hydrogen exchanger 1 (NHE1) protein. NHE1 is a transmembrane transporter that maintains intracellular pH (pHi) by exchanging internal protons for external sodium ions. In various cancers, including kinase-mutated acute myeloid leukemia (AML), NHE1 is overexpressed or hyper-activated, leading to an alkaline intracellular environment that promotes cell survival, proliferation, and resistance to therapy. By knocking down NHE1 expression, this shRNA induces intracellular acidification and triggers apoptosis. Developed by researchers at the University of Hong Kong, this modality has demonstrated the ability to suppress leukemic growth in vivo and synergize with tyrosine kinase inhibitors and chemotherapy by altering the transmembrane pH gradient.
