ShotCAR-GRP78 is a preclinical-stage chimeric antigen receptor (CAR) cell therapy being developed by Hauul Bio for the treatment of various solid tumors, including triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and colorectal cancer (CRC). The therapy utilizes Hauul Bio's proprietary ShotCAR platform to engineer immune cells that target Glucose-Regulated Protein 78 (GRP78), also known as HSPA5. GRP78 is a molecular chaperone typically resident in the endoplasmic reticulum, but it is frequently overexpressed and translocated to the cell surface in a wide range of cancers, where it promotes tumor cell survival, migration, and resistance to therapy. By specifically targeting surface-expressed GRP78, ShotCAR-GRP78 aims to induce potent anti-tumor activity while minimizing toxicity to normal cells where GRP78 remains intracellular.
