Overview
SHP656 is a long-acting, polysialylated recombinant Factor VIII (rFVIII) drug candidate that was developed by Shire (now part of Takeda) in collaboration with Xenetic Biosciences. The molecule utilizes Xenetic's proprietary **PolyXen** platform technology, which conjugates polysialic acid (PSA) to the therapeutic protein to extend its half-life and improve its pharmacokinetic profile. The primary goal of the program was to develop a once-weekly treatment for **hemophilia A**. Despite demonstrating an extended half-life and a favorable safety profile in clinical testing, Shire terminated the development of SHP656 in May 2017 after a Phase 1/2 trial failed to meet the pre-defined once-weekly dosing criterion.
