shPAR1 is an experimental short hairpin RNA (shRNA) designed to silence the expression of Protease-Activated Receptor 1 (PAR1), also known as coagulation factor II receptor (F2R). In the context of pancreatic ductal adenocarcinoma (PDAC), research indicates that PAR1 signaling plays a complex role; while stromal PAR1 promotes tumor progression, tumor cell-specific PAR1 expression is associated with the maintenance of a mesenchymal phenotype. Silencing PAR1 via shRNA in tumor cells has been shown to increase epithelial characteristics and proliferative index while decreasing metastatic potential in murine models. This research was primarily conducted at the Academic Medical Center of the University of Amsterdam.
