Overview

shPRMT9 is a short hairpin RNA (shRNA) designed to silence the expression of Protein Arginine Methyltransferase 9 (PRMT9) through RNA interference. In the context of oncology research, specifically Acute Myeloid Leukemia (AML), shPRMT9 is used to demonstrate that PRMT9 is essential for the maintenance of leukemia stem cells (LSCs). By knocking down PRMT9, researchers observed impaired survival of primary AML cells and delayed leukemogenesis in mouse models. PRMT9 normally catalyzes the methylation of PABP1 at the R493 residue, a process that promotes protein synthesis and AML viability; shPRMT9 disrupts this oncogenic signaling pathway.
