shPTGDS is a short hairpin RNA (shRNA) designed to specifically knock down the expression of prostaglandin D2 synthase (PTGDS). In diabetic kidney disease (DKD), PTGDS is significantly upregulated in podocytes under high glucose conditions. This upregulation, mediated by STAT1, leads to the nuclear translocation of NF-κB, which accelerates the transcription of senescence-associated secretory phenotypes (SASPs) factors and promotes podocyte injury. By knocking down PTGDS, shPTGDS aims to attenuate high glucose-induced podocyte senescence and injury, thereby alleviating podocyte and glomerular injury. This therapeutic approach has been investigated in podocyte-specific AAV9 harboring shPTGDS mice.
