SHR-1222 is a novel humanized monoclonal antibody targeting sclerostin (SOST), a protein that inhibits bone formation. By inhibiting sclerostin, SHR-1222 promotes bone formation and decreases bone resorption, making it a promising therapeutic candidate for osteoporosis treatment. It has been shown in clinical studies to increase bone mineral density (BMD) notably at the lumbar spine and modulate biomarkers related to bone metabolism such as increasing N-terminal propeptide of type 1 procollagen, osteocalcin, and bone-specific alkaline phosphatase while decreasing β-C-telopeptide. The drug is administered subcutaneously and has demonstrated dose-dependent pharmacokinetics with good tolerability in phase I and II clinical trials conducted primarily in China. Developed by Jiangsu Hengrui Medicine Co., SHR-1222 is currently in phase II development for osteoporosis.
