SHR-1701 + platinum-doublet chemotherapy is an investigational combination regimen being evaluated for the treatment of unresectable stage III non-small cell lung cancer (NSCLC). SHR-1701 is a bifunctional fusion protein (biologic) that simultaneously targets two immune-suppressive pathways: Programmed Death-Ligand 1 (PD-L1) and Transforming Growth Factor-beta Receptor II (TGF-βRII). By blocking PD-L1, it restores T-cell anti-tumor activity, while its TGF-β trap component inhibits TGF-β signaling to address immune exclusion and fibrosis in the tumor microenvironment. The platinum-doublet chemotherapy component (typically cisplatin or carboplatin paired with a second agent like pemetrexed) provides direct cytotoxic effects and may enhance tumor antigenicity. This combination is being studied as induction and consolidation therapy to improve outcomes in patients undergoing concurrent chemoradiotherapy.
