SHR-1701 + S-1 + oxaliplatin is a combination therapy investigated for advanced or metastatic solid tumors, notably gastrointestinal cancers. SHR-1701 is a bifunctional fusion protein that simultaneously targets programmed death ligand 1 (PD-L1) and transforming growth factor-beta (TGF-β), blocking immunosuppression and modulating the tumor microenvironment. S-1 is an oral fluoropyrimidine consisting of tegafur (a prodrug of 5-fluorouracil), gimeracil, and potassium oteracil; it inhibits thymidylate synthase and therefore DNA synthesis. Oxaliplatin is a platinum-based chemotherapeutic that causes DNA crosslinking, resulting in cytotoxicity and apoptosis. This triple regimen is under clinical evaluation as first-line or perioperative treatment, particularly in gastrointestinal cancers such as metastatic colorectal and gastric cancer.
