SHR-1920 is a humanized IgG1 monoclonal antibody that specifically targets trophoblast cell-surface antigen 2 (Trop-2). Developed by Hengrui Medicine, it serves as the targeting antibody component of the antibody-drug conjugate (ADC) SHR-A1921. SHR-1920 binds to Trop-2 with nanomolar affinity in humans and rhesus monkeys but does not show cross-reactivity with rat or mouse Trop-2. Trop-2 is a cell-surface glycoprotein overexpressed in a variety of epithelial carcinomas, including prostate, lung, and breast cancers, making it a valuable target for directed cytotoxic therapy. In the context of SHR-A1921, SHR-1920 is conjugated to a topoisomerase I inhibitor payload to selectively deliver the toxin to tumor cells.
