SHR-2524 is an investigational humanized monoclonal antibody that targets the T-cell immunoreceptor with Ig and ITIM domains (TIGIT), an inhibitory immune checkpoint protein expressed on the surface of T cells and Natural Killer (NK) cells. Developed by Suzhou Suncadia Biopharmaceuticals, a subsidiary of Jiangsu Hengrui Pharmaceuticals, SHR-2524 is designed to bind to TIGIT and block its interaction with its primary ligands, CD155 (PVR) and CD112 (PVRL2), which are often overexpressed on tumor cells and antigen-presenting cells. By inhibiting this pathway, SHR-2524 aims to reverse immune suppression and restore the anti-tumor activity of effector immune cells. In clinical development, it is being evaluated for the treatment of advanced solid tumors, including hepatocellular carcinoma (HCC), where it is frequently studied in combination with other therapies such as the anti-VEGF antibody bevacizumab or the PD-1 inhibitor camrelizumab to achieve synergistic anti-cancer effects.
