SHR-4298 is an investigative antibody-drug conjugate (ADC) developed by Jiangsu Hengrui Medicine and its subsidiary, Suzhou Suncadia Biopharmaceuticals. The molecule is designed to target the Six-transmembrane epithelial antigen of prostate 2 (STEAP2), a cell surface protein that is significantly overexpressed in several malignancies, most notably prostate cancer, while maintaining restricted expression in healthy tissues. SHR-4298 consists of a humanized anti-STEAP2 monoclonal antibody conjugated to a potent topoisomerase I inhibitor payload (likely an exatecan derivative) via a cleavable linker. Upon binding to the STEAP2 antigen on the surface of tumor cells, the ADC is internalized through the endocytic pathway. Inside the cell, the linker is cleaved, releasing the cytotoxic payload which then inhibits DNA topoisomerase I, leading to double-strand DNA breaks and subsequent apoptosis. SHR-4298 is currently being evaluated in Phase I/II clinical trials for the treatment of various advanced malignant solid tumors.
