SHR-4602 is a next-generation HER2-targeting antibody-drug conjugate (ADC) designed to deliver a potent cytotoxin specifically to HER2-expressing tumor cells. It utilizes an enzyme-cleavable linker technology and incorporates the amino acid sequence of pertuzumab with ER300 (an eribulin derivative) as its payload. This design aims to overcome resistance seen in tumors treated with earlier HER2 ADCs, including those using topoisomerase I inhibitor payloads. Preclinical studies have demonstrated strong anti-tumor activity both *in vitro* and *in vivo*, particularly against tumors resistant to other anti-HER2 therapies. The drug is being developed primarily for advanced malignant solid tumors that are HER2-positive or have HER2 alterations.
