Overview

SHR-4849 is a novel antibody-drug conjugate (ADC) that targets Delta-like protein 3 (DLL3), a protein highly expressed in small cell lung cancer (SCLC) and neuroendocrine tumors (NETs). The drug consists of an anti-DLL3 antibody linked to a topoisomerase I inhibitor payload, enabling targeted delivery of cytotoxic activity to DLL3-expressing tumor cells. Preclinical studies have demonstrated strong antitumor activity, including tumor regression as monotherapy and significant bystander effects against DLL3 low-expressing cells. In Phase 1 clinical trials for advanced solid tumors, particularly SCLC and NETs, SHR-4849 has shown encouraging early efficacy signals with partial responses observed in the majority of evaluable SCLC patients. The safety profile is considered manageable, with most treatment-related adverse events being Grade 1 or 2. The drug is being developed through a collaboration between Hengrui Pharma and IDEAYA Biosciences[1][2][4][5][6][7].
