Overview

SHR-7012 is a humanized monoclonal antibody developed by Hengrui Pharmaceutical that targets Blood Dendritic Cell Antigen 2 (BDCA2), also known as CD303. BDCA2 is a receptor exclusively expressed on the surface of plasmacytoid dendritic cells (pDCs), which are the primary producers of Type I interferons (IFN-I). By binding to BDCA2, SHR-7012 induces receptor internalization and suppresses the activation of pDCs, thereby inhibiting the production of IFN-I and other inflammatory mediators. This mechanism is designed to treat autoimmune conditions characterized by an overactive interferon pathway, such as cutaneous lupus erythematosus (CLE) and systemic lupus erythematosus (SLE). The drug is currently undergoing early-phase clinical evaluation to assess its safety, pharmacokinetics, and pharmacodynamics.
