SHR-A1811 + camrelizumab + famitinib is a combination therapy being evaluated in the FUTURE-SUPER platform trial (NCT05656131) led by Fudan University for the treatment of refractory triple-negative breast cancer (TNBC). The combination consists of three distinct agents: SHR-A1811, a next-generation antibody-drug conjugate (ADC) targeting HER2; camrelizumab, a humanized monoclonal antibody targeting the PD-1 receptor; and famitinib, an oral multi-targeted tyrosine kinase inhibitor (TKI). SHR-A1811 delivers a cytotoxic topoisomerase I inhibitor payload directly to HER2-expressing tumor cells. Camrelizumab acts as a checkpoint inhibitor to restore T-cell mediated anti-tumor immunity. Famitinib inhibits angiogenesis and tumor cell proliferation by targeting VEGFR2, VEGFR3, PDGFR, and c-kit. This triplet regimen aims to overcome therapeutic resistance in TNBC by simultaneously targeting tumor cell survival, immune evasion, and the tumor microenvironment.
