Overview

**SHR-A1811 + paclitaxel albumin-bound** is an investigational combination of two antineoplastic drugs, composed of:\n- **SHR-A1811**: an antibody-drug conjugate targeting human epidermal growth factor receptor 2 (HER2). It consists of trastuzumab (the anti-HER2 antibody), a cleavable linker, and a novel topoisomerase I inhibitor payload (SHR169265). SHR-A1811 is designed to deliver cytotoxic payload selectively to HER2-expressing or mutated tumor cells, resulting in DNA damage, apoptosis, and antitumor efficacy. The drug was developed to improve potency and decrease toxicity via optimal payload permeability, drug-to-antibody ratio (DAR=6), and plasma stability[1][3][5][7].\n- **Paclitaxel albumin-bound** (nab-paclitaxel): a formulation of paclitaxel bound to albumin nanoparticles, facilitating better tumor delivery and allowing higher dosing. Paclitaxel inhibits microtubule disassembly, disrupting mitotic and cellular function, leading to cell cycle arrest and apoptosis.
