Overview

This combination therapy is a multi-drug regimen being investigated for the first-line treatment of advanced gastric or gastroesophageal junction adenocarcinoma. It consists of SHR1701, a bifunctional fusion protein that simultaneously targets Programmed cell death 1 ligand 1 (PD-L1) and Transforming growth factor beta (TGF-β); SHR2554, a small-molecule inhibitor of Histone-lysine N-methyltransferase EZH2; and the CAPOX chemotherapy regimen, which includes capecitabine and oxaliplatin. The rationale for this combination is to leverage the synergistic effects of immune checkpoint blockade, TGF-β inhibition, and epigenetic modulation to overcome primary and acquired resistance to standard immunotherapy and chemotherapy in gastric cancer patients.
