Overview

**SHR3680 + metformin** is an unbranded combination of two drugs: SHR3680 (rezvilutamide), a novel non-steroidal antiandrogen that antagonizes the androgen receptor used primarily in prostate cancer, and metformin, a widely used biguanide antidiabetic that acts mainly via activation of AMP-activated protein kinase (AMPK) and inhibition of hepatic gluconeogenesis. SHR3680 binds the androgen receptor and prevents androgen-induced gene expression, thereby inhibiting the growth of androgen-dependent prostate cancer cells[2][6]. Metformin decreases blood glucose levels by decreasing hepatic glucose production and improving insulin sensitivity. There is strong preclinical and rational support for combining antiandrogens and metformin in prostate cancer and metabolic comorbidity, but as of now, there is no clinical trial or marketed product specifically for the SHR3680 + metformin combination.
