Overview

SHR6390 + letrozole + anastrozole is an experimental combination regimen investigated for the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. SHR6390, also known as dalpiciclib, is a selective cyclin-dependent kinase (CDK) 4/6 inhibitor. Letrozole and anastrozole are nonsteroidal aromatase inhibitors that decrease estrogen synthesis, which is essential for hormone-sensitive breast cancer growth. This combination leverages both targeted cell cycle inhibition (via CDK4/6 blockade) and suppression of estrogen production, offering a comprehensive approach to hormonal breast cancer therapy. The combination has been evaluated in early-phase (IB/II) and phase III clinical trials, showing promising antitumor activity and acceptable safety profiles in this patient population[1][3][5][7][9].
