SHRN1126

SHRN1126 is an orally administered, steroidal selective estrogen receptor degrader (SERD) and antagonist developed for the treatment of advanced ER-positive/HER2-negative metastatic breast cancer. It fully blocks estrogen-induced transcriptional activity without any agonist effects, distinguishing it from some other SERDs. In preclinical models, SHRN1126 demonstrated strong efficacy in suppressing tumor growth—including in both ESR1 wild-type and mutant breast cancer—and showed synergy when combined with other endocrine therapies. The drug has superior pharmacokinetic properties such as oral bioavailability and favorable performance compared to current therapies like fulvestrant and elacestrant. Preclinical toxicology studies indicate an excellent safety profile with no genotoxic or cardiovascular concerns[2][4].
