Overview

shRNA AKR1B10 is a short hairpin RNA-based therapeutic candidate designed to silence the expression of Aldo-keto reductase family 1B10 (AKR1B10). AKR1B10 is an enzyme involved in the metabolism of lipid substrates, including the reduction of farnesal/geranylgeranial to farnesol/geranylgeraniol, which is a critical step for protein prenylation and KRAS activity. In preclinical models of pancreatic cancer, knockdown of AKR1B10 using shRNA has been shown to inhibit cell growth, invasion, and migration, as well as suppress tumor growth in vivo by down-regulating active KRAS and its downstream signaling pathways, including phosphorylated C-Raf, ERK, and MEK1/2.
