Overview

shRNA-DNMT3b is a short hairpin RNA (shRNA) designed to target and silence the expression of DNA methyltransferase 3b (DNMT3b). In MYC-driven malignancies such as T-cell acute lymphoblastic leukemia (T-ALL) and Burkitt's lymphoma (BL), the MYC oncogene directly upregulates DNMT3b, which in turn maintains aberrant DNA methylation patterns at CpG islands near gene promoters. This epigenetic regulation leads to the silencing of genes involved in differentiation and apoptosis while promoting tumor cell proliferation. By utilizing RNA interference to knock down DNMT3b levels, shRNA-DNMT3b disrupts these tumor-maintenance programs, leading to the upregulation of genes responsible for differentiation and development and the downregulation of those controlling proliferation and survival. It is primarily used as a research tool to validate DNMT3b as a therapeutic target in MYC-dependent cancers.
