Overview

shRNA-GPC3 (short hairpin RNA targeting Glypican-3) is a gene-silencing tool used in molecular biology and oncology research to knock down the expression of Glypican-3 (GPC3). GPC3 is a cell-surface heparan sulfate proteoglycan that is highly expressed in embryonic tissues and various cancers, such as hepatocellular carcinoma (HCC) and lung squamous cell carcinoma (LUSC), but is minimally expressed in normal adult tissues. In laboratory models, shRNA-GPC3 is typically delivered via viral vectors (such as lentivirus) or plasmid transfection to study the functional role of GPC3 in tumor cell proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT).
