shRNA-Notch3 is a short hairpin RNA (shRNA) therapeutic candidate designed to knockdown the expression of the Notch3 receptor. In multiple myeloma (MM), Notch3 signaling between myeloma cells and osteocytes in the bone marrow niche promotes tumor proliferation and bone destruction. By reducing the protein levels of the active Notch3 intracellular domain (NICD3), this shRNA approach inhibits downstream Notch target genes and cyclinD1, leading to decreased MM cell proliferation and increased apoptosis. Preclinical studies in mouse models have demonstrated that Notch3 knockdown significantly reduces tumor burden and osteolytic lesions, suggesting that targeting Notch3 in the tumor microenvironment is a viable strategy for treating MM and its associated bone disease.
