shRNA PD-L1 refers to short hairpin RNA designed to target and silence the expression of programmed death-ligand 1 (PD-L1). PD-L1 is a key immune checkpoint protein that is frequently upregulated on the surface of tumor cells, allowing them to evade immune detection by binding to PD-1 on T cells. In addition to its role in immune evasion, tumor-intrinsic PD-L1 has been shown to promote tumor-initiating cell generation, epithelial-to-mesenchymal transition (EMT), cell proliferation, and migration. Silencing PD-L1 using shRNA is widely utilized in preclinical research to investigate these cell-autonomous roles of PD-L1 and to evaluate the therapeutic potential of gene-silencing approaches in various cancers, including triple-negative breast cancer, melanoma, and ovarian cancer.
