Overview

shRNA targeting FASN is an experimental RNA interference (RNAi) therapeutic modality designed to silence the expression of the Fatty Acid Synthase (FASN) enzyme. FASN is a multi-enzyme protein that catalyzes de novo fatty acid synthesis and is frequently overexpressed in various malignancies, including glioblastoma (GBM), where it contributes to tumor growth and immune evasion. Research presented at SNO 2021 indicates that FASN-mediated lipid synthesis hampers anti-cancer immunity, particularly following radiation therapy (RT). By utilizing an inducible shRNA to knockdown FASN in murine GBM cell lines (such as GL261 and CT2A), researchers demonstrated reduced tumor growth, increased secretion of type I interferons (IFN-beta) and CXCL10, and enhanced infiltration of CD8+ T cells and CD11c+ dendritic cells in orthotopic mouse models. This approach suggests that targeting FASN can sensitize tumors to radiotherapy and promote anti-cancer immunity.
