Overview

**SI-B001 + paclitaxel** is a combination therapy comprised of SI-B001, a novel bispecific monoclonal antibody targeting epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 3 (HER3), and paclitaxel, a microtubule inhibitor. SI-B001 (also known as izalontamab) blocks both EGFR and HER3, key drivers in tumor proliferation, with demonstrated antitumor activity in EGFR-driven epithelial cancers. Paclitaxel disrupts cell division by stabilizing microtubules. This combination is primarily being investigated for recurrent and metastatic head and neck squamous cell carcinoma (HNSCC), especially after progression following anti-PD-1 immunotherapy and platinum-based chemotherapy. In phase II studies, this combination has shown improved objective response rate and disease control rate compared to SI-B001 monotherapy, with manageable toxicity[1][3][4].
