Overview

SI-B001 + SI-B003 + chemotherapy is a combination regimen under clinical investigation for the treatment of recurrent, locally advanced, or metastatic head and neck squamous cell carcinoma and non-small cell lung cancer. SI-B001 (izalontamab) is a tetravalent bispecific monoclonal antibody targeting EGFR and HER3, designed to block signaling from both receptors and mediate immune cell effector functions against cancer cells[4]. SI-B003 (danvilostomig) is a tetravalent bispecific antibody targeting PD-1 and CTLA-4, aiming to restore anti-tumor activity of exhausted tumor-specific T cells through immune checkpoint inhibition[4]. Chemotherapy is administered concomitantly (typically platinum-based regimens[1][6]). The combination seeks to directly inhibit growth factor receptor-driven proliferation while enhancing anti-tumor immunity.
