SI-B003 (danvilostomig) is a tetravalent bispecific monoclonal antibody that targets both PD-1 (programmed cell death protein 1) and CTLA-4 (cytotoxic T-lymphocyte-associated protein 4), two key immune checkpoint receptors on T cells. By simultaneously blocking these inhibitory pathways, SI-B003 aims to restore and enhance the anti-tumor activity of exhausted tumor-specific T cells. The molecule is designed with two binding domains for each target, leveraging bi-valency and bi-specificity to improve immune cell function against cancer. It is being developed primarily for the treatment of advanced or metastatic solid tumors, including patients who have progressed on prior PD-1/L1 immunotherapy[1][5][8]. Clinical trials have demonstrated a tolerable safety profile and preliminary anti-tumor activity in this patient population[1][3].
