Drug intelligence / Profile preview
si-ssCPLC is a preclinical small interfering RNA (siRNA) therapeutic designed to target and silence the long non-coding RNA (lncRNA) CPLC (also known as LINC00893). In inflammatory bowel disease (IBD), CPLC is upregulated in lesions and acts as a molecular sponge that binds TAF1A, disrupting the TAF1A-ZBTB34 complex and releasing ZBTB34 to translocate to the nucleus. This activates the transcription of the glycolytic enzyme LDHA, driving pathological glycolytic reprogramming, lactate accumulation, and mucosal barrier damage. Delivered orally via lesion-specific, chondroitin sulfate-modified curcumin nanobottles (CCNB), si-ssCPLC knocks down CPLC expression, thereby reversing pathological glycolysis, restoring intestinal barrier proteins, and reducing inflammatory cytokines.
