SIAH2 siRNA is a small interfering RNA designed to silence the expression of the Seven in Absentia homolog 2 (SIAH2) gene. SIAH2 is a RING finger E3 ubiquitin ligase that serves as a critical signaling node downstream of the EGFR/HER2/K-RAS pathway. By inhibiting SIAH2, this therapy induces oxidative stress, mitochondrial dysfunction, and apoptosis in cancer cells, particularly those addicted to oncogenic K-RAS signaling. Preclinical studies have demonstrated that SIAH2 knockdown leads to increased reactive oxygen species (ROS) production and decreased phosphorylation of ERK1/2 and AKT, effectively combating invasive and metastatic cancer lines such as non-small cell lung adenocarcinoma and breast cancer.
